چکیده
In the field of studies of bacterial toxicity, acute toxicity induced by bacterial agents, particularly bacteria of the genus Shigella sp. with Shigella dysenteriae at the top, has been relevant since the Shigella dysenteriae has been highly recognized for its production of one of the major virulence factors for humans such as Shiga toxin (Stxs). Shiga toxins are a family of genetically and functionally related cytotoxic proteins expressed by the enteric pathogens Shigella dysenteriae serotype 1 and certain serotypes of Escherichia coli (Lee et al., 2010). Recently, numerous studies have shown that Shiga toxins induce apoptosis through different mechanisms in different cell types including human epithelial cells, endothelial cells and neurons (Tesh, 2010). The use of human of renal cortical epithelial cells and a renal carcinoma cell line to investigate the cytotoxic effects of Stxs has shown the induction of apoptosis in both cell types as evidenced by nuclear fragmentation, DNA laddering, and the presence of terminal deoxynucleotidyltransferase-mediated dUTP-biotin nick end labelling (TUNEL)-positive cells (Lentz et al., 2011). Stxq1-mediated apoptosis of monocytic THP-1 cells involved a novel pathway using components of both the intrinsic and extrinsic pathways of apoptosis (Lee et al., 2005). In the case of the excretion product of Shigella dysenteriae (SdyEP) as inducer of apoptosis, this process has been highlighted by the description of morphological signals, indicative of cell death Int. J. Morphol., 31(4):1175-1180, 2013.

Dual AO/EB Staining to Detect Apoptosis in Osteosarcoma Cells Compared with Flow Cytometry
BACKGROUND The aim of this study was to evaluate the ability of dual acridine orange/ethidium bromide (AO/EB) staining to detect tumor cell apoptosis. According to apoptosis-associated changes of cell membranes during the process of apoptosis, a clear distinction is made between normal cells, early and late apoptotic cells, and necrotic cells. MATERIAL AND METHODS We cultured human osteosarco...
متن کاملCritical Evaluation of Techniques to Detect and Measure Cell Death – Study in a Model of UV Radiation of the Leukaemic Cell Line HL60
The reliability of eight distinct methods (Giemsa staining, trypan blue exclusion, acridine orange/ethidium bromide (AO/EB) double staining for fluorescence microscopy and flow cytometry, propidium iodide (PI) staining, annexin V assay, TUNEL assay and DNA ladder) for detection and quantification of cell death (apoptosis and necrosis) was evaluated and compared. Each of these methods detects di...
متن کاملApoptotic and Necrotic Effects of Pectic Acid on Rat Pituitary GH3/B6 Tumor Cells
Background: Pectin is composed of complex polysaccharides that can inhibit cancer metastasis and proliferation with no evidence of toxicity. In the present study, the apoptotic and necrotic effects of pectic acid (PA) on the rat pituitary GH3/B6 tumor cells has been investigated. Methods: GH3/B6 cells were cultured in the Ham’s F12 medium enriched with 15% horse serum and 2.5% fetal bovine seru...

This study was conducted to investigate the modulating effects of green tea polyphenols on genotoxic damage and apoptotic activity induced by hexavalent chromium [Cr (VI)] in CD-1 mice. Animals were divided into the following groups: (i) injected with vehicle; (ii) treated with green tea polyphenols (30 mg/kg) via gavage; (iii) injected with CrO3 (20 mg/kg) intraperitoneally; (iv) treated with ...

Nowadays natural compounds are increasingly used in various fields since they cause fewer adverse side effects than their chemical counterparts. In addition, preparation and production of natural compounds are comparatively simple. The present study aimed to evaluate the anti-cancer effects of brittle star methanol extract on MCF-7 cells. Anti-cancer effects of brittle star methanol extract wer...
